P3 and NIa-Pro of Turnip Mosaic Virus Are Independent Elicitors of Superinfection Exclusion

چکیده

Superinfection exclusion (SIE) is an antagonistic interaction between identical or closely related viruses in host cells. Previous studies by us and others led to the hypothesis that SIE was elicited one more proteins encoded genomes of primary viruses. Here, we tested this using Turnip mosaic virus (TuMV), a member genus Potyvirus family Potyviridae, with significant economic consequences. To end, individual TuMV-encoded were transiently expressed cells Nicotiana benthamiana leaves, followed challenging them modified TuMV expressing green fluorescent protein (TuMV-GFP). Three days after TuMV-GFP delivery, these examined for replication-dependent expression GFP. Cells P1, HC-Pro, 6K1, CI, 6K2, NIa-VPg, NIb, CP permitted efficient GFP, suggesting failed block replication superinfecting TuMV-GFP. By contrast, N. P3 NIa-Pro did not express visible GFP fluorescence, both could elicit potent against The elicitor activity further confirmed their heterologous from different potyvirus, potato A (PVA). Plants systemically infected PVA variants blocked subsequent infection +1-frameshift mutation cistrons facilitated superinfection TuMV-GFP, proteins, but RNA, are involved activity. Additionally, deletion mutagenesis identified amino acids 3 200 352 40 181 242 as essential elicitation. Collectively, our study demonstrates encodes two spatially separated act independently exert on TuMV. These results lay foundation mechanistic interrogations virus.

The Nuclear Inclusion a (NIa) Protease of Turnip Mosaic Virus (TuMV) Cleaves Amyloid-β

BACKGROUND The nuclear inclusion a (NIa) protease of turnip mosaic virus (TuMV) is responsible for the processing of the viral polyprotein into functional proteins. NIa was previously shown to possess a relatively strict substrate specificity with a preference for Val-Xaa-His-Gln↓, with the scissile bond located after Gln. The presence of the same consensus sequence, Val(12)-His-His-Gln(15), ne...

gD-Independent Superinfection Exclusion of Alphaherpesviruses.

UNLABELLED Many viruses have the capacity to prevent a cell from being infected by a second virus, often termed superinfection exclusion. Alphaherpesviruses, including the human pathogen herpes simplex virus 1 (HSV-1) and the animal herpesvirus pseudorabies virus (PRV), encode a membrane-bound glycoprotein, gD, that can interfere with subsequent virion entry. We sought to characterize the timin...

The NIa-Pro protein of Turnip mosaic virus improves growth and reproduction of the aphid vector, Myzus persicae (green peach aphid).

Many plant viruses depend on aphids and other phloem-feeding insects for transmission within and among host plants. Thus, viruses may promote their own transmission by manipulating plant physiology to attract aphids and increase aphid reproduction. Consistent with this hypothesis, Myzus persicae (green peach aphids) prefer to settle on Nicotiana benthamiana infected with Turnip mosaic virus (Tu...

Equilibrium sedimentation of turnip yellow mosaic virus.

Sedimentation equilibrium was achieved with turnip yellow mosaic virus at low speeds (600 rpm) in a magnetic ultracentrifuge. The experiments were carried out in the newly installed constant-speed rotor, equipped with automatic control and an electromagnetic drive. A particle mass of 5.55 x 10(6) daltons was calculated for the virus at vanishing concentrations, in essential agreement with the e...

Replication and encapsidation of recombinant Turnip yellow mosaic virus RNA.

Turnip yellow mosaic virus (TYMV) is a positive strand RNA virus that infects mainly Cruciferae plants. In this study, the TYMV genome was modified by inserting an extra subgenomic RNA promoter and a multiple cloning site. This modified TYMV was introduced into Nicotiana benthamiana using a Agrobacterium-mediated T-DNA transfer system (agroinfiltration). When a gene encoding beta-glucuronidase ...
